Safranal Inhibits HeLa Cell Viability by Perturbing the Reassembly Potential of Microtubules

🎀🎀Saffron, a spice from Crocus sativus, has been known for its health benefits and medicinal properties. Safranal is a component of saffron and is known for its antioxidant and anticancer properties. In this study, we elucidated a possible tubulin-targeted antiproliferative mechanism of action of safranal.

🏜️🏜️In vitro, the compound perturbed secondary structure of tubulin without altering net microtubule polymer mass. It inhibited HeLa cell viability in a concentration-dependent manner, with minimal damage to cellular microtubules. However, it strongly inhibited recovery of microtubule network after cold-induced disassembly, indicating its ability to interfere with the nucleation potential of tubulin.

🏜️🏜️Further, as the acetylation pattern of the safranal-treated microtubules revealed, unlike many tubulin-targeted agents, the compound did not appear to induce persistent stabilization of microtubules. Our data shows an unusual, tubulin-targeted antiproliferative mechanism of safranal.

Efficacy of Crocus sativus (saffron) in treatment of major depressive disorder associated with post-menopausal hot flashes: a double-blind, randomized, placebo-controlled trial
💯💯Purpose: Due to concerns regarding the side effects of hormone therapy, many studies have focused on the development of non-hormonal agents for treatment of hot flashes. The aim of this study was to evaluate the efficacy and safety of saffron (stigma of Crocus sativus) in treatment of major depressive disorder associated with post-menopausal hot flashes.
💯💯Methods: Sixty women with post-menopausal hot flashes participated in this study. The patients randomly received either saffron (30 mg/day, 15 mg twice per day) or placebo for 6 weeks. The patients were assessed using the Hot Flash-Related Daily Interference Scale (HFRDIS), Hamilton Depression Rating Scale (HDRS) and the adverse event checklist at baseline and also at the second, fourth, and sixth weeks of the study.
💯💯Results: Fifty-six patients completed the trial. Baseline characteristics of the participants did not differ significantly between the two groups. General linear model repeated measures demonstrated significant effect for time × treatment interaction on the HFRDIS score [F (3, 162) = 10.41, p = 0.0001] and HDRS score [F (3, 162) = 5.48, p = 0.001]. Frequency of adverse events was not significantly different between the two groups.
💯💯Conclusions: Results from this study revealed that saffron is a safe and effective treatment in improving hot flashes and depressive symptoms in post-menopausal healthy women. On the other hand, saffron, with fewer side effects, may provide a non-hormonal and alternative herbal medicine option in treatment of women with hot flashes.

1️⃣1️⃣ Anxiety and depression have high prevalence in the general population, affecting millions of people worldwide, but there is still a need for effective and safe treatments. Nutritional supplements have recently received a lot of attention, particularly saffron. Thus, several pre-clinical studies support a beneficial role for bioactive compounds, such as saffron, in anxiety and depression.

⏳⏳ Here we used an animal model of depression based on social isolation to assess the effects of affron®, a standardized saffron extract containing ≥3.5% of total bioactive compounds safranal and crocin isomers. Saffron was administered both through the oral and the intraperitoneal routes, and several tasks related to anxiety and depression, such as the elevated plus maze, the forced swimming test or the sucrose preference test, were assessed. These tasks model key features of depressive states and anxious states relating to fear, behavioral despair or anhedonia, the lack of motivation and/or pleasure from everyday activities, respectively.

1️⃣1️⃣Animals receiving oral saffron displayed behaviors congruent with improvements in their anxious/depressive state, showing the enhanced consumption of a sweet solution, as well as an increase in certain escape responses in the forced swimming test. Our data support a beneficial role for oral saffron in anxious/depressive states.

However, if we consider only the legal definition, they can only be referred to as new foods, medical foods, dietary supplements and herbs or medicinal plants [148]. Interestingly, the journal Nature Biotechnology was already describing the concept of “agriceuticals” in 1999.
🥊🥊The safety of saffron as a spice has been established by its use as a food additive to provide color and flavor over centuries, while traditionally extracts and tinctures of saffron have been used in the treatment of diseases with no toxic side effects .
🥊🥊An extract from the 1987 European Commission monograph about saffron discussed the dangers of dosages in food: “to a daily maximum dose of 1.5 grams there has not been any risk documented. Lethal dose is 20 g and the abortive dose, 10 g, because as such it was employed in the past due to its stimulating action on the smooth muscle of the uterus. 5 g daily dose can already cause intoxication accompanied by vomiting, bloody diarrhea, hematuria, skin hemorrhages in nose, lips and eyelids, vertigo and dulling. The skin and mucous membranes take a yellowish colour similar to jaundice”.
High doses of saffron should be avoided during pregnancy.
🥊🥊In a sub-acute assessment, it was seen that saffron is a safe spice with no toxic effects on the liver, although the administration of high doses of saffron to mothers could damage the kidneys of the newborn .

How do Saffron stigmas affect Inflammatory Bowel Diseases?

🎓🎓Kawabata et al. reported that 4 weeks crocin feeding was able to inhibit Dextran Sulfate Sodium (DSS)-induced colitis and decrease tumor necrosis factor α expression, interleukin- (IL-) 1β, IL-6, interferon γ, NF-κB, cyclooxygenase-2, and inducible nitric oxide synthase in the colorectal mucosa and increased Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) expression .

🎓🎓Results suggested that crocin suppressed chemically induced colitis and colitis-associated colon carcinogenesis in mice, at least partly by inhibiting inflammation and the expression of certain pro-inflammatory cytokines and inducible inflammatory enzyme .

☀☀In a recent study, crocin demonstrated anti-ulcerogenic and coloprotective effects. The therapeutic impact was mediated primarily via enhancement of colon Nrf2 content by 211% in protective protocol and by 350% in curative protocol and HO-1 signaling by 49% in protective protocol and, 288% in the curative protocol. Enhancement of Nrf2 and HO-1 signaling and down-regulation of caspase-3 activity are believed to underly the observed therapeutic effect .

☀☀Therefore, crocin could be a candidate for both the prevention of colitis and inflammation-associated colon cancer.

📌 Types of Saffron

While Iran is the top saffron producer and their production is known for the best quality in the world. However, there are different kinds of Saffron, dividing into four major types, each with different quality. These four types are Poshal, Sargol, Negin, and Super Negin.

1. Poshal: The cheapest and least quality type of Saffron is known as Poshal. This Saffron is from yellow parts of the stigma which contains no value, taste, and aroma.
2. Sargol: In Farsi it literally means "Top of the flower" which derives from the very tip of the saffron thread. It has a specific, powerful aroma more than other types. It is also known as high quality since it does not contain any Yellow or Orange threads. However, during the harvest, and separation process, Sargol saffron often contains crumbs and broken threads.
3. Negin: This type of Saffron is longer in length but contains some yellow and orange portions. However, the aroma, quality, and purity are still great but the color is not perfect due to some portions of yellow and orange threads, which makes this type less quality than others.
4. Super Negin: This type is the most expensive but rare trim of Saffron. It is known not only for the most potent but also for its aesthetically pleasing features. Super Negin threads are longer and contain no yellow or orange threads. The threads are cut symmetrically and contain no crumbs. When comparing all the trims of Saffron, Super Negin is the best considering the quality, color, and taste.
