A crime scene is often silent. There may be no eyewitnesses, no fingerprints, and no obvious clues. Yet a tiny drop of blood, a strand of hair with its root, or even a few skin cells left behind can tell an extraordinary story.
Hidden inside those cells is DNAโa molecular record unique to almost every individual. But comparing an entire human genome for every investigation would be far too slow and computationally expensive. Scientists needed a faster approach. That solution became the Combined DNA Index System (CODIS).

Instead of reading an entire genome, CODIS examines 20 standardized genetic markers (19 STR loci and the Amelogenin marker). The combination of these markers forms a DNA profile that is extraordinarily unlikely to be shared by two unrelated individualsโmaking it as unique as a fingerprint, but written in DNA.

From a tiny biological sample to a database match, the process follows five key steps:
1. DNA is extracted from blood, saliva, hair roots, or other biological evidence.
2. The CODIS genetic markers are amplified using PCR.
3. Capillary electrophoresis determines the allele pattern at each marker.
4. These patterns are converted into a numerical DNA profile.
5. The profile is searched against millions of records in CODIS to identify potential matches.
