Wolfram syndrome is a rare genetic disorder that affects approximately 3,000 people living in the U.S. with no approved treatment options.

Common manifestations of Wolfram syndrome include insulin-requiring diabetes mellitus, vision loss, diabetes insipidus, hearing loss, neurogenic bladder, impaired balance and coordination, and difficulty breathing that can progress to respiratory failure. We’re pleased to share that Week 24 and Week 48 results from our Phase 2 open-label HELIOS clinical trial in Wolfram syndrome have been published in the Journal of Clinical Investigation. These findings indicate continued improvement in pancreatic function and visual acuity stabilization over 48 weeks, as well as a generally well-tolerated safety profile. Calpain-2 (CAPN2) is an important contributor to axonal degeneration, or nerve fiber damage, which plays a critical role in ALS progression.

Research suggests that when calpain-2 becomes overactive, it can interfere with normal cell function and weaken the nerve fibers that carry signals throughout the body.

Post-bariatric hypoglycemia (PBH) is a rare, chronic condition with no FDA-approved treatments. Among the roughly two million people who have had Roux-en-Y gastric bypass (RYGB) or sleeve gastrectomy in the past decade in the U.S., an estimated 160,000 are living with PBH today.

The condition can cause debilitating low blood sugar episodes when the brain does not receive enough glucose. PBH can be difficult to diagnose as symptoms, such as brain fog, fatigue, sweating, blurred vision, and dizziness, can be non-specific, vary in severity, and/or be mistaken for dumping syndrome.
