Amyotrophic lateral sclerosis ( ) is an adult-onset neurodegenerative disorder characterized by progressive muscle weakness due to degeneration of upper motor neurons in the brain and lower motor neurons in the brainstem and spinal cord.

Diagnosis relies on clinical criteria with mixed upper and lower motor neuron features, and is often supported by electromyography and genetic testing, which may direct treatment.

A prospective UK Biobank cohort study assessed the interplay of modifiable and genetic risk factors for across age groups. The study found that the association of modifiable risk factors—including smoking, excessive alcohol intake, unhealthy diet, physical inactivity, overweight, diabetes, and pancreatitis—with pancreatic cancer was strongest among adults younger than 60 years.
