قد تتحول البكتيريا موجبة الجرام الى سالبة الجرام
وذلك لعدةظروف اهمها:
🔸تقدم المزارع الموجبة بالسن فتتحول لسالبة.
🔸تأثر الموجبة بارتفاع درجة حموضة البيئة.
🔸استخدام الكحول لفترة طويلة.
🔸سحق البكتيريا الموجبة ببرادة زجاج او المنيوم مما يؤدي الى تهشم الجدار الخلوي فتفقد البكتيريا المتهشمة ايجابيتها.
🔸عندما تتعامل الخلايا مع انزيمribonuclease الذي يذيب الحمض الرايبوزي النووي ribo nucleic acid ويذيب املاح المغناسيوم معه.
ونضيف الي الاتي *↓↓
🔹زيادة فترة التثبيت للشريحة على اللهب اثناء عمل الفيلم يكسر الجدار الخلوي للبكتيريا
🔹استخدام صبغات منتهية الاستخدام
وخاصة الــ. Iodine المسئول عن تثبيت الصبغه في البكتيريا الموجبة
