The melatonin molecule has a very high safety profile, with only a few difficult-to-quantify subjective side effects occasionally being mentioned, e.g., dizziness, sleepiness, and headache. Melatonin has been given at a dose of 1000 mg daily for 30 days with no evidence of toxicity, and many other authors have noted its minimal or negligible side effects when ingested daily by humans. When used to defer or treat diseases, the human-equivalent dose allometrically determined on the basis of results from animal studies is estimated to be 1.0–1.5 mg per kg of body weight. A major contributor to these challenges is sarcopenia—the age-related loss of muscle mass. By age 70, individuals typically lose about 30% of their muscle, resulting in diminished leg strength, slower reflexes, and impaired balance, all of which heighten the risk of falls.
