Q1. Mention the principle of Sysmex's 3-part differential haematology analyser.
A1. The analyser uses two main measurement principles:
a. Electrical impedance (aperture impedance) principle:
RBCs, WBCs and platelets suspended in a conductive electrolyte solution pass individually through a small aperture between two electrodes. As each cell passes through the aperture, it causes a temporary increase in electrical resistance, producing an electrical pulse.
• The number of pulses corresponds to the number of cells counted.
• The amplitude of each pulse is proportional to the volume of the cell.
• The cells are therefore classified according to their size and counted from a measured volume of blood.
b. Photometric/spectrophotometric principle for haemoglobin:
Haemoglobin is measured photometrically after the RBCs are haemolysed in the WBC/Hb dilution. The photometric system includes a light source, optical system/cuvette and photodetector.
